Creatine-Facilitated Prevention of Stress Induced by Violated Circadian Rhythm

We studied the protective role of creatine on the changes caused by extended violated circadian rhythm in white rat hippocampus. We established that daily injection of 140 mg/kg creatine during 30 days improved physiological and certain characteristics caused by stress induced by violated circadian rhythm among rats. In particular, the treatment reduced the quantity of NO, H2O2 and Ca2+ ions that had previously risen as a result of the stress. The results also showed an increase in the formerly lowered activity of antioxidant enzymes, particularly of SOD and catalase. We assume that the reason in the heightened activity of these enzymes in rat hyppocampal cells may lie in the creatine-induced activation of energy metabolism and certain intracellular signaling routes, followed by heightening of synthesizing reactions and increase in the quantity of antioxidant enzymes. The process can be conditioned by creatine’s impact on the NMDA receptor, which consequently reduces previously heightened Ca2+ quantity and neutralizes its cytotoxic effect, boosting enzyme synthesis and their content, as well as other processes.
